Examination of the role of ADP-ribosylation factor and phospholipase D activation in regulated exocytosis in chromaffin and PC12 cells

The possible role of ADP-ribosylation factor (ARF)-activated and constitutive phospholipase D (PLD) activity in regulated exocytosis of preformed secretory granules in adrenal chromaffin and PC12 cells was examined. With use of digitonin-permeabilised cells, the effect of GTP analogues and exogenous ARF1 on PLD activity was determined. No evidence was seen for ARF-stimulated PLD activity in these cell types. Exocytosis from cytosol-depleted permeabilised chromaffin cells was not increased by adding recombinant nonmyristoylated or myristoylated ARF1, and exocytosis from both cell types was resistant to brefeldin A (BFA). Addition of bacterial PLD with demonstrably high activity in permeabilised chromaffin cells did not increase exocytosis in cytosol-depleted chromaffin cells. Diversion of PLD activity from production of phosphatidic acid (PA) due to the presence of 4% ethanol did not inhibit exocytosis triggered by Ca2+ or poorly hydrolysable GTP analogues in permeabilised chromaffin or PC12 cells. These results indicate that exocytosis in these cell types does not appear to require a BFA-sensitive ARF and the triggering of exocytosis does not require PLD activity and formation of PA. These findings rule out a general requirement for PLD activity during regulated exocytosis.     

The incidence and severity of rheumatoid arthritis, results from a county register in Oslo, Norway

OBJECTIVE: To examine the incidence of rheumatoid arthritis (RA) in the community of Oslo, Norway; and to link the incidence to measures of disease severity. METHODS: A register for RA in the county of Oslo was validated to be 85% complete for patients between 20 and 79 years of age. The incidence of RA was calculated from this register for patients with disease onset 1988-1993 (n=550). Data on health related quality of life [Modified Health Assessment Questionnaire (MHAQ), Arthritis Impact Measurement Scale 2 (AIMS2), pain, fatigue] were obtained from a postal survey (response rate 73%). RESULTS: The overall annual incidence of RA over a 6 year period was 25.7/100,000 (females 36.7, males 13.8). The incidence increased with age, from 7.8 in the age group 20-29 to 61.0 between 70 and 79 years. After 5 year followup a clinically important effect on physical function (MHAQ > 1.5) was seen in 38% of the patients, on social functioning (AIMS2 social > 4) in 50%, on mental distress (AIMS2 affect > 4) in 27%, on pain (VAS > 40 mm) in 35%, and on fatigue (VAS > 40 mm) in 41%. CONCLUSION: The incidence of RA was 25.7/100,000 in the population of Oslo. After 5 years, 40-50% had clinically important changes in health status. The annual disease incidence with clinically important effect on physical function after 5 years was 10/100,000 individuals at risk.     

Investigation of the oxidation of o-Xylene over a Vanadia/Titania catalyst by means of the TAP reactor

The oxidation of o-xylene over a commercial V₂O₅/TiO₂ (anatase) catalyst was studied in the TAP (Temporal Analysis of Products) reactor system. The transient method with high time resolution has led to new insights into the reaction mechanism. The selective oxidation is shown to consist of a sequence of steps, involving intermediates which desorb as o-toluolaldehyde and phtalide. The key reaction step in the non-selective oxidation is shown to be a decar-bonylation of a surface species, leading to a surface benzoate. The kinetics of the elementary steps in the reaction were derived from a quantitative analysis of the transient responses on o-xylene pulses.     

The self-serving bias in relational context.

This article examined the impact of relationship closeness on the self-serving bias (SSB). Members of relationally distant dyads working on interdependent-outcomes tasks manifested the SSB: They took credit for dyadic success but blamed the partner for dyadic failure. However, members of relationally close dyads did not manifest the SSB: They did not take more credit than their partner for dyadic success and did not blame the partner more than the self for dyadic failure. This gracious attributional pattern of relationally close dyed members is due, at least in part, to formation of a favorable impression of the partner. Interbunch extinction measurement at the BNL AGS for the KOPIO experiment

The KOPIO experiment at the BNL AGS required an extracted proton beam in which a debunched, coasting beam is forced between empty longitudinal RF buckets to form microbunches. The goal of the measurement described here was to obtain a low background determination of the fraction of protons coming from the AGS slowly extracted beam at times between microbunches (interbunch extinction). The effect on interbunch extinction of variations in the RF cavity voltage, the momentum dispersion of the beam and the main guide field voltage-ripple was studied. It was found that there exists a broad range of operating parameters that could allow the AGS to produce microbunches with the interbunch extinction better than the KOPIO experiment requirement. Results obtained in this study gave interbunch extinctions better than 10⁻⁵ (1%) of KOPIO's upper limit.
